I am thrilled to share the latest progress on our groundbreaking gene therapy facility in Braine-l'Alleud, Wallonia, Belgium. The exterior of the facility is now complete, and we are currently moving forward with the “Fit Out” activities. The first equipment is delivered and already in place. This marks a significant milestone in our journey towards revolutionizing disease treatment through advanced gene therapy.

Why is this project important? For ��ֳ�, it means opening new opportunities to discover and develop cures for the people we serve. For me, it's deeply personal. People living with severe diseases are at the heart of everything we do, and every morning, I’m motivated by the thought that this facility could one day help my loved ones.

Beyond the personal and corporate impact, this state-of-the-art facility, representing a substantial investment in Belgium, is designed to be both innovative and environmentally sustainable. It will significantly enhance our development and manufacturing capabilities, reinforcing our existing teams in Braine-l’Alleud (Belgium), Leuven (Belgium), Boston (USA), and Durham (UK).

Our new facility will allow us to have complete ownership of our chemistry, manufacturing, and controls (CMC) capabilities. This will enable us to be more agile in responding to new trends and opportunities, and more scalable to meet the unmet needs of patients. The efficient clinical manufacturing capability will ensure we maintain high standards of speed, scalability, and quality as we bring our gene therapy research projects to patients.

Building Gene Therapy Skills

In addition to the physical construction of the facility, we are also committed to developing our skills and expertise in gene therapy and more largely bio-related fields. This field offers a wealth of opportunities for both current employees and new hires. We are excited to announce that the novel facility will create over 100 new, highly skilled jobs. We are actively seeking talented, inspired, and passionate individuals who are eager to join us in improving lives through innovative therapies.

From unlocking the secrets of chemicals, cultivating the promises of pharmaceuticals and expanding into biologics, peptides and gene therapy, our researchers at ��ֳ� are on a mission to continuously innovate. To do that, we are harnessing today’s new frontiers in science and technology and strengthening our capabilities to discover and develop new medicines. So, it should come as no surprise that we are utilising some unique disruptive technology to help strengthen our gene therapy activities.

We recently signed an agreement with CEVEC Pharmaceuticals to evaluate and gain access to their ELEVECTA® technology. ELEVECTA® technology may enable ��ֳ� to develop a scalable, robust and efficient manufacturing of gene therapy vectors. Up until now, manufacturing of adeno-associated viral vectors (AAVs) has been an expensive and challenging process usually requiring multiple batches of transient transfection of cells lines and large quantities of cGMP-quality plasmids. The CEVEC technology overcomes these limitations based on producer cell-lines which have all the necessary elements for AAV production integrated in one cell. This enables superior, efficient, high-performance, industry scale AAV production from one cell line with consistent quality.

At ��ֳ� we have world class discovery, research, development and manufacturing facilities and to maintain this position we will continuously monitor new and disruptive technologies in the pursuit of innovation.

This is great news as it advances our ability to develop novel treatments using state of the art gene therapy technology.

��ֳ� has a vision to move from symptomatic treatments to disease modification and eventually towards a cure. Adeno-associated virus (AAV) mediated gene therapy offers to deliver that potential and drive a fundamental change in how diseases are treated with the ability to remove or add disease-related proteins with a single treatment. A vast array of severe diseases are amenable to gene therapy and ��ֳ� is embracing this modality to expand its capabilities and ultimately transform the lives of patients with severe diseases.

Handl Therapeutics BV is a Leuven-based, Belgium born, transformative gene therapy company with a vision to deploy the power of disease modifying in vivo gene therapy to treat complex neurodegenerative diseases. Operating in a highly collaborative manner, the company built a strong international network to access global capabilities and expertise. To this end, it combines state of the art technology platforms and scientific advances licenced from KU Leuven (Belgium), Centre for Applied Medical Research (CIMA Universidad de Navarra, Spain), University of Chile (Chile) and King’s College London (UK) to address unmet medical needs.

To transform the lives of people living with severe diseases in immunology and neurology, we too must transform. We must keep innovating, reinventing ourselves and adapting to the trends around the world. We want to tap into the latest scientific advances and best talent. Only then can we develop the best life-changing therapies to help our patients live their lives to their fullest.

Over the years, ��ֳ� has moved from small molecules to biologics and peptides. We recently announced our first steps into gene therapy. Gene therapy will fundamentally change how we treat diseases, moving us from symptomatic treatments to disease modification and eventually towards a cure. It offers enormous potential for massively transforming the lives of people living with severe diseases.

Transforming our Braine l’Alleud site (South of Brussels)

In addition to moving into gene therapy, we’re also transforming our sites, organization, and infrastructure for the future. Take the multi-product biological manufacturing facility we’re building on our Braine l’Alleud site in Wallonia, Belgium.

The Braine l’Alleud site was formerly a high-volume, lower-value site mainly focused on small-molecule and chemistry. Then, ten years ago, we started investing in our first BioPilot plant, with chemistry, manufacturing and control capabilities for biologics. Our new manufacturing facility represents a significant new step on that journey, transforming Braine l’Alleud into high-tech and high-value site.

Construction of the large-scale facility started in 2020, and we expect the new facility to be operational in 2024. The plant itself will be highly modern, green by design and with a significant digital integration. It will provide the facilities we need at the scale we need to launch and supply the medicines we currently have in clinical development – and we have a rich pipeline of biologics that we plan to launch between now and 2025.

Building our expertise and talent

I am incredibly proud of how our dedicated staff have made our journey towards biologics manufacturing possible, investing their time in developing new competencies and enriching the diversity of thoughts and approaches to increase our agility. Simultaneously, we also need to continue building skills and expertise as biologics and dealing with high-value materials requires specific competencies, complex data analysis and robust quality management.

When it comes to biologics, innovation and sciences unveil an unlimited landscape , as does digitalization. It’s an area with a lot of opportunities. And maximizing the value we gain from data and taking that process to the next stage is a huge opportunity before us today.

We expect to create more than 150 new, high-skilled jobs once our new manufacturing plant in Braine l’Alleud becomes fully operational. That’s why we’re on the lookout for talented, inspired, daring and engaged individuals who, like us, have an ambition to create value for patients.
